The activation site is where the binding of an aminoacid to the 3´amino acid pocket in tRNA happens. The enzym catalyzes the reaction between these two to create an "esterbindning" between the CCA in tRNA's amino acid seat/pocket to the C in the aminoacid. More specfically, it is adenine that directly binds to the tRNA. The aminoacyl-tRNA is then moved to the editing site where it gets checked that the right aminoacid has bound to the tRNA to see if its anticodons are compatible with the mRNAs codons.
