The activation site of the Aminoacyl-tRNA synthetase is responsible for adding an amino acid to the tRNA that carries the right anticodon. The binding of the amino acid and tRNA is then controlled by the editing site. If the wrong amino acid is added, the binding between the amino acid and tRNA is cleaved, and a new amino acid gets added by the activation site to the tRNA.
The activation site is where the binding of an aminoacid to the 3´amino acid pocket in tRNA happens. The enzym catalyzes the reaction between these two to create an "esterbindning" between the CCA in tRNA's amino acid seat/pocket to the C in the aminoacid. More specfically, it is adenine that directly binds to the tRNA. The aminoacyl-tRNA is then moved to the editing site where it gets checked that the right aminoacid has bound to the tRNA to see if its anticodons are compatible with the mRNAs codons.
